how can we come to know that weather it is do follow or not ?

You can know do-follow or no-follow by following method.

Right click on specific link and and go to properties option and see the "will open in" field Link properties option. if you see same window in this option then this the link is do-follow and if you find no-follow in that option then it would no-follow.

You can also use a do follow search for only do follow sites with links.

Directory submission and also forum posting in do follow site can also of help in link building. Blog commenting is really helpful but a lot of blogger doesnt approved commenting anymore.

A very special tactic I use are Podcasts.
YOu can use odiogo to convert written text to audio and submit your RSS feed to Podcast Directories
